Analysis of Genetic Diversity of Onion Germplasm Using RAPD

Abstract
Genetic variation and relationships of onion germplasm by RAPD-PCR analysis were investigated. In the test of 120 primers with 58 onion lines and cultivars collected from local and abroad, eight primers turned out to be useful for further RAPD analysis. These primers provided a total of 71 bands, consisting of 68 polymorphic and three monomorphic ones. The polymorphic bands were subjected to phylogenetic analysis using NTSYS and PAUP computer programs. In the NTSYS analysis, similarity coefficient of 58 onion lines and cultivars ranged from 0 to 0.9179 and similarity coefficient of 27 varieties with obvious origin ranged from 0.0306 to 0.9032. The dendrogram of 58 varieties obtained by NTSYS analysis resulted in 15 groups, but overall groupings were inconsistent with phenotypes (e.g., color and shape) of bulb classified at the collection site. In the PAUP analysis, genetic divergence of 58 onion lines and cultivars ranged from 0.042 to 0.532 and that of 27 lines and cultivars with obvious origin ranged from 0.042 to 0.394. Phylogenetic analysis using PAUP revealed 13 groups with 58 lines and cultivars and nine groups with 27 lines and cultivars, but these were inconsistent with phenotypes of bulb at the collection site, as in the case of NTSYS analysis.
